Description:
This method calculates plasma membrane potential (ΔψP) time course based on previously known baseline potential and kP using the FLIPR Membrane Potential Kit from Molecular Devices (aka PMPI, plasma membrane potential indicator).
Required experimental protocol: baseline, followed by any time lapse. The final complete depolarization is measured in a separate sample starting from an identical baseline.
This calibration is recommended only for averaged fluorescence of many cells and not for individual cells, because individual cell fP0 value is variable.
