DHE rate of ethidium formation normalized to dihydroethidium

Parameters:
Name # Type Description
Channel for ethidium (OH-ethidium) 1 integer Channel number for the numerator in the fluorescence ratio calculation
Channel for dihydroethidium (blue fluorescence) 2 integer Channel number for the denominator in the fluorescence ratio calculation
Background Level, Percentile 3 real Background is calculated as frame-by-frame mean of those pixels that are the darker than this percentile of image histogram for the duration of the entire recording.
Mask sensitivity (0.2-1.5) 4 real Sensitivity factor, typically 0.2-1.5
Ranges where to calculate rates 5 string If left empty, all time points are shown, or give ranges to calculate means e.g. 1-3;7-9. Try also copy/paste marks in the plot window context menu.
Description:
Calculates average normalized ethidium (or OH-ethidium) fluorescence rate in all cells in the view field after stabilization of images, background subtraction and masking background regions. To eliminate background, the “Mean of pixels below percentile of max projection” with the percentile given at “Background level” is used.
In the Excel Data Window output the rate of ethidium formation is given in the column Ch1_ROI1 in fluorescence units / sec and the normalization factor is in the column Ch2_ROI1. Calculate Ch1_ROI1/Ch2_ROI1 for normalized rates (1/sec).
