PMPI Short Calibration between known baseline and separately measured fP0 ( IAPMPIShortBaselinefP0 )
Parameters:
Name |
Short Name |
Type |
Description |
Range definitions in time courses: |
rangedef |
string |
Defines whether the ranges given below refer to frames or seconds in the recordings |
Baseline Range: |
BaselineRange |
string |
Defines the range of the baseline (in the above defined time units). Longer baseline results more accurate calibration. |
ΔψP at baseline (mV): |
PMP0 |
real |
Previously measured value of ΔψP at baseline. |
kP: |
kP |
real |
Previously measured value of kp |
fP0: |
fP0 |
real |
Separately measured value of fp0 |
Description:
This method calculates plasma membrane potential (ΔψP) time course based on previously known baseline potential and kP.
Required experimental protocol: baseline, followed by any time lapse. The final complete depolarization is measured in a separate sample starting from an identical baseline.
This calibration is recommended only for averaged fluorescence of many cells and not for individual cells, because individual cell fP0 value is variable.
